Summary

I am a member of the nPOD-Virus Group. I have applied immuno-EM using the Tokyuasu method to detect VP1 and dsRNA in CBV infected human cell lines, as well as mouse pancreas with acute and persistent EV infections, the latter obtained using terminally deleted virus provided by Dr. Nora Chapman then used to generate infected mice by Marc Horwitz. Based on these encouraging results, Alberto Pugliese asked that I examine a small number of pancreata from organ donors with type 1 diabetes (T1D), to obtain proof of principle that this method can detect natural EV infections in humans. As presented at the annual nPOD meeting and in the preliminary data presented in this application, our results provide immuno-EM evidence of EV infection in nPOD T1D pancreata. With this application, I intend to expand on these preliminary observations, as detailed in the specific aims. Based on this promising results we propose to examine a larger number of nPOD diabetic and non-diabetic pancreas tissue samples, to address novel questions about virus infections in the T1D pancreas at the ultrastructural levels.
