Enteroviruses (EVs), belonging to picornaviruses, contain several medically important viruses, such as polioviruses (PVs), coxsackieviruses, numbered EVs, and rhinoviruses. Many findings about entry and uncoating of PVs were originally thought to be shared by all EVs, but recent research has revealed that, despite very similar structures and genomes, there are interesting differences to be discovered among them. EVs make use of the general receptor-recycling pathways to gain entry to cell, but EVs can also trigger their own entry pathway as has been shown, especially for members of the EVB group. Recent studies on physiologically relevant factors on uncoating and advances in the methods to study uncoating have also shed light on this perhaps least well-known aspect during EV entry. In this chapter, accumulated information on both entry and uncoating will be reviewed in detail.
